Highly pure alpha-hederin from Hedera helix leaves isolated by centrifugal partition chromatography

Introduction: Hedera helix is a species of the Araliaceae family, which contains flavonoids, coumarins, phenolic acids, sterols, alkaloids, anthocyanins, and triterpene saponins. α-Hederin saponin compound H. that possesses many potential biological activities. In previous studies, several methods were used to isolate purify α-hederin from ivy leaves. However, loss obtained during solid chromatography purification was relatively high, it difficult obtain with high purity. this research, centrifugal partition (CPC) method established for isolation directly ethanol crude extract leaves Da Lat, Lam Dong province, Vietnam. Methods: The separation carried out by applying PLC 2250 system (Gilson Inc., France) coupled an SCPC-250 column (Armen Instrument, France). CPC run, two-phase solvent consisting n-hexane–ethyl acetate–methanol–water (2:3:2:3, v/v) in ascending mode (the lower phase stationary phase), rotation speed 1600 rpm flow rate 8.0 mL/min, used. Results: purity isolated 95.7%, as confirmed HPLC-DAD at 210 nm. structure purified determined 1H-NMR 13C-NMR. yield 12.7%. Conclusion: This investigation might be reference large-scale high-purity
